红细胞生成素（erythropoietin，EPO）对红细胞生成的特异性刺激作用的细胞因子。红细胞生成素是一种糖蛋白，在胎儿体内由肾脏及肝脏产生，而在成人体内主要由肾脏产生。目前已知人红细胞生成素有两种存在形式，即人EPO-α及人EPO-β，二者氨基酸组成及顺序相同，都含有165个氨基酸残基。分子量、等电点及生物活性也都类似，差别在于二者的糖型组成不同，EPO-α含有较多的N-乙酰氨基葡萄糖和N-乙酰神经氨酸，总的含糖量也较EPO-β高。红细胞生成素-α是调节红细胞增殖、成熟和分化的一种造血生长因子。

 人红细胞生成素-α重组蛋白是在CHO中国仓鼠卵巢细胞中重组表达的，是一条单链的多肽链，包含有165个氨基酸，计算分子量为21，000Da，由于发生糖基化修饰而导致实际分子量为30，400 Da。该人红细胞生成素-α重组蛋白最终由标准的色谱技术纯化出。

【EPO a】Recombinant Human Erythropoietin-alpha 重组人红细胞生成素-α  细胞因子实验操作须知：

溶解：提供的人红细胞生成素-α(EPO a)产品为过滤灭菌的白色冻干粉末；该冻干粉末中 人红细胞生成素-α、柠檬酸钠、NaCl、柠檬酸的比率分别为：1mg、0.58mg、0.58mg、0.006 mg。我们建议加无菌的18MΩ-cm水以配制工作储存液到100ug/ml，后继实验过程中可根据实验要求进一步稀释。

保存：人红细胞生成素-α(EPO a)冻干粉末在室温可保存3周，或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上；如果想延长保存时间，建议分装（加载体蛋白如0.1% HSA或 BSA）后置于-18℃冻存。请避免反复冻融。

Introduction 

This gene is a member of the EPO/TPO family and encodes a secreted, glycosylated cytokine composed of four alpha helical bundles. The protein is found in the plasma and regulates red cell production by promoting erythroid differentiation and initiating hemoglobin synthesis. This protein also has neuroprotective activity against a variety of potential brain injuries and antiapoptotic functions in several tissue types.

Description 

Erythropoietin-alpha Human Recombinant is produced in Chinese hamster ovary (CHO) cells by recombinant DNA technology is a single, polypeptide chain containing 166 amino acids and having a predicted molecular mass of 21,000 Dalton and apparent glycosylated molecular mass of 36-40kDa. EPO-a is purified by proprietary chromatographic techniques.

Physical Appearance 

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

Each mg of lyophilized powder contains 0.59 mg sodium citrate, 0.58 mg sodium chloride and 0.006 mg citric acid.

Solubility 

It is recommended to reconstitute the lyophilized Epoetin-a in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability 

Lyophilized Erythropoietin-a although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution EPO-alpha should be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.
