白细胞介素-1（interleukin-1,IL-1）是在感染和炎症状态下，由多种细胞产生的、有多方面生物学功能的细胞因子，其生物学作用十分广泛，主要包括参与介导炎症反应、调节免疫、调节机体代谢。IL-1分子重量为17.5KD，有两种类型：IL-1α和IL-1β，主要以IL-1β为主。IL-1受体（interleukin-1 receptor，IL-1R）包括Ⅰ型和Ⅱ型，此外还存在一种内源性IL-1受体拮抗剂 （interleukin-1 receptor antagonist，IL-1ra或IL-1γ），IL-1ra可以与IL-1受体结合，但是不引起相应的作用。炎症情况下,粒细胞巨噬细胞集落刺激因子(GM-CSF) 水平升高，它也能促进IL-1 的上调作用,增加IL-1β的合成;另外关节滑膜细胞也可自分泌IL-1β。
人白介素-1β重组蛋白是在大肠杆菌中重组表达的，是一条非糖基化的多肽链，包含有153个氨基酸，分子量为17000Da。该人白介素-1β重组蛋白最终由标准的色谱技术纯化出。

溶解：提供的人白介素-1β (IL 1 beta)产品为过滤灭菌的白色冻干粉末；该冻干粉末是从含1mg/ml人白介素-1β (IL 1 beta)的50mM 磷酸缓冲液（pH 7.1）和150mM NaCl溶液中过滤灭菌并冻干产生的。我们建议加无菌的18MΩ-cm水以配制工作储存液到100ug/ml，后继实验过程中可根据实验要求进一步稀释。

保存：人白介素-1β (IL 1 beta)冻干粉末在室温可保存3周，或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天；如果想延长保存时间，建议分装（加载体蛋白如0.1% HSA或 BSA）后置于-18℃冻存。请避免反复冻融。

Introduction 

Interleukin-1b is produced by activated macrophages, IL-1B stimulates thymocyte proliferation by inducing il-2 release, b-cell maturation and proliferation, and fibroblast growth factor activity. IL1B proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.

Description 

Interleukin-1 beta Human Recombinant produced in E.Coli is a non-glycosylated, Polypeptide chain containing 153 amino acids and having a molecular mass of 17000 Dalton.
The IL-1b is purified by proprietary chromatographic techniques.

Physical Appearance 

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

The protein was lyophilized from a concentrated (1mg/ml) sterile solution containing 50mM Phosphate buffer pH=7.1 and 150mM NaCl.

Solubility 

It is recommended to reconstitute the lyophilized Interleukin-1b in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability  （稳定性）

Lyophilized Interleukin-1b although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution IL1b should be stored at 4°C between 2-7 days and for future use below -18°C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.

Amino acid sequence （氨基酸序列）

The sequence of the first five N-terminal amino acids was determined and was found to be Ala-Pro-Val-Arg-Ser.

Biological Activity  （生物活性）

The specific activity as determined in the test of augmentation of lymphocyte proliferation assay using mouse thymus was found to be 200,000,000 IU/mg.

Protein content  （蛋白质含量）

Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.631 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of IL-1 beta Recombinant as a Reference Standard.
