粒细胞集落刺激因子(granulocyte colony stimulating factor,G-CSF)基因全长2.5kb，包括5个外显子和4个内含子，人G-CSF基因位于17号染色体，与小鼠G-CSF基因约有73％同源性，并具有相互交叉的生物学活性，且与IL-6无论在基因水平以及氨基酸水平上都有很高同源性，包括外显子、内含子组成，Cys数目，两对二硫键位置以及分子的三级结构等。G-CSF主要作用于中性粒细胞系造血细胞的增殖、 分化和活化。在体外G-CSF刺激骨髓造血祖细胞中中性粒细胞集落的形成，延长成熟中性粒细胞的存活时间，活化中性粒细胞如促进ADCC，超氧阴离子的产生和碱性磷酸酶的合成。G-CSF还具有对人粒细胞、单核细胞、成纤维细胞、平滑肌细胞以及成肌纤维细胞的趋化作用。肿瘤患者注射G-CSF后可提高血循环中中性粒细胞的水平，这种作用可能与缩短某些骨髓细胞进入S期的时间以及增加生成粒细胞的祖细胞数量有关。

溶解：提供的小鼠粒细胞集落刺激因子(mG CSF)产品为过滤灭菌的白色冻干粉末；该冻干粉末是从含小鼠粒细胞集落刺激因子(mG CSF)的溶液（不含添加剂）中冻干产生的。我们建议加无菌的18MΩ-cm水以配制工作储存液到100ug/ml，后继实验过程中可根据实验要求进一步稀释。
保存：小鼠粒细胞集落刺激因子(mG CSF)冻干粉末在室温可保存3周，或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上；如果想延长保存时间，建议分装（加载体蛋白如0.1% HSA或 BSA）后置于-18℃冻存。请避免反复冻融。

Introduction 

Granulocyte Colony Stimulating Factor is a growth factor and/or cytokine produced by the endothelium, macrophages and a number of other immune cells. GCSF stimulates the bone marrow to produce granulocytes and also to stimulate the survival, proliferation, differentiation and function of neutrophil granulocyte progenator cells and mature neutrophils.

Description 

Granulocyte Colony Stimulating Factor Mouse Recombinant produced in E.coli is a single, non-glycosylated, polypeptide chain containing 179 amino acids and having a molecular mass of 19kDa.
G-CSF is purified by proprietary chromatographic techniques.

GCSF was lyophilized with no additives.

Solubility  （溶解性）

It is recommended to reconstitute the lyophilized Granulocyte Colony Stimulating Factor in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability  （稳定性）

Lyophilized Granulocyte Colony Stimulating Factor although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution GCSF should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

The ED50 range=0.01-0.03 ng/ml corresponding to a Specific Activity of 33,4000,000-100,000,000IU/mg, determined by the dose-dependant proliferation of mouse NFS- 60cells. The optimal concentration for each specific application should be determined by an initial dose-response assay.
